In Blue Exorcist: The Blue Night Saga episode 2, the story begins with Rin Okumura witnessing a discussion between Mephisto and Shiro Fujimoto, with the latter demanding to become an Exwire, so he can eventually earn his freedom.
However, the demon emphasizes how becoming a Paladin won't matter much because he is going to continue as a prisoner, but Shiro continues to challenge him.
The next portion of the episode focused on Mephisto revealing to Rin that the Asylum, which would go on to become the Exorcist Academy in the future, was created so he could come up with experiments. He created multiple potential vessels for Lucifer's new body since the latter is currently suffering greatly, and being taken care of, while receiving a potential immortality elixir.
Blue Exorcist: The Blue Night Saga episode 2 shows a completely dejected Rin as he discovers that Mephisto has been their enemy, especially considering the impact that seeing Lucifer once again had on him. This is, along with the introduction of Satan in the previous episode, the biggest revelation that the protagonist has had thus far.

The second portion of Blue Exorcist: The Blue Night Saga episode 2 moves four years into the future in the Exorcist Academy, showing how Shiro Fujimoto has grown older and stronger. He still has to go through different experiments, with the episode depicting many of the individuals made through this process dying and Shiro vowing to stay alive.
The final part of the episode has Rin unable to keep watching, and moves a bit forward into the future as he sees Yuri Egin and several other potential Exorcists dealing with the threat of an Ent.
Eventually, the creature is taken care of by Shiro, who makes a massive display of power and also shows a strong disgust for Yuri's compassion for demons, with the flames of Satan also shown in the forest they are in.
Final thoughts
The episode had the hefty revelation of the reason behind Mephisto's experiments and the state of Lucifer, much to the protagonist's disgust. It also gave the audience a greater understanding of Shiro Fujimoto's struggles in his youth and the many ordeals he had to go through in order to stay alive.
