No, Inojin Yamanaka does not die in Boruto: Two Blue Vortex manga as Himawari Uzumaki manages to save him in time.

The manga's previous chapter saw Himawari Uzumaki attack Jura in hopes of stopping his assault. However, the God Tree manages to easily counter her and break her leg. Right after, he trapped her within a Wood-Style entrapment jutsu.

Upon witnessing this, Inojin gets enraged as he attacks Jura with one of his Super Beast Scroll jutsu. When that failed, Inojin himself changed toward the God Tree to fight him. Unfortunately, he too was instantly knocked down. Right after, Jura impaled Inojin by creating a tree from right below Inojin's torso. These developments led fans to believe that Inoijin wasn't going to survive in Chapter 11.

Fortunately, the developments seen in Boruto: Two Blue Vortex Chapter 11 were favorable for Inojin. After Himawari Uzumaki was able to access Kurama's chakra, she obtained the tailed beast's super regenerative abilities. With that, Himawari healed her broken leg in no time, following which, she shifted her focus to Inojin.

As seen at the end of the previous chapter, Himawari's hair had started looking similar to that of Kurama's tails. Using the same, Himawari lifted Inojin up from the top of the tree and instantly healed him using her regenerative powers. With that, Inojin's wound began closing up. Hence, Himawari saved Inojin by using Kurama's regenerative powers on him.

That said, Himawari herself wasn't certain if she had healed Inojin completely. Hence, she asked Shikadai Nara and Chocho Akimichi to take Inojin to a hospital immediately. Thus, while the alleged manga spoilers did not show Inojin waking up after getting impaled by God Tree Jura, it effectively confirmed that his life was no longer in danger.

Even if, through some complications, Himawari might have failed in healing Inojin entirely, Shikadai and Chocho taking him to the hospital should see him get saved. This is because the Konoha Medical Department was headed by Sakura Haruno, who was recognized as one of the greatest medical-nin in the world.
