Quantrill attended Trinity College School in Port Hope, Ontario, where he was a four-time varsity letterman in baseball, hockey, and volleyball. He continued his athletic career at Stanford University, playing baseball for the Stanford Cardinal. In the 2016 MLB Draft, he was selected as the eighth overall by the San Diego Padres.

After playing through the minor leagues, Cal Quantrill made his Major League debut on May 1, 2019, with the San Diego Padres, where he played until 2020. On August 31, 2020, he was traded to the Cleveland Guardians.
