Duke Jr. began his basketball career at Classical High School in his hometown, where he averaged 15 points per game and led his team to a Division 1 state title during his junior season. He later transferred to Cushing Academy in Ashburnham, Massachusetts, where he led his high school team to a 17-5 record and to the NEPSAC AA Semifinals in 2018 during his senior season. Duke Jr. attended Providence College in 2018, where he was named to the Preseason All-BIG EAST First Team, the Maui Invitational All-Tournament Team, the Second Team All-BIG EAST, and the USBWA All-District 1 Team in the 2020-21 season. Internationally, Duke Jr. represented the United States men's basketball team in the 2019 Pan American Games in Peru, where the team secured the bronze medal.

Duke Jr. remained undrafted in the 2021 NBA Draft and was signed with the Brooklyn Nets on August 8, 2021. He signed with the Long Island Nets in the NBA G League after signing on October 16, 2021. He was named to the NBA G League Next-Up Game in 2023, the All-NBA G League First Team in 2023, and the Second-Team All-Big East in 2021. In the 2023 NBA G League, he played for the Delaware Blue Coats.
