Agugua-Hamilton's coaching journey began at her alma mater VCU where she spent a season as a graduate assistant. In the next two seasons, she was named the assistant coach and led VCU to its first NCAA Tournament appearance. She also worked at Indiana for two straight seasons from 2009 to 2011. Agugua-Hamilton also was the assistant and recruiting coordinator for two seasons where she led the program to a WNIT berth during her second year.

Before earning her first head coaching role at Missouri State, Hamilton spent six years with Michigan State as an assistant and associate head coach. She helped the Spartans to four NCAA Tournament appearances and a Big Ten regular-season title. She was named the head coach of the Missouri State Lady Bears in 2019, a position she held for the next three seasons. She led the Lady Bears to two MVC regular-season titles and an overall 74-15 record. The two-time MVC Coach of the Year was named coach of Virginia’s women's basketball program in 2022.
