Here's a compilation of every one of those key moments you might have missed about what the game was trying to hint at:
#1 V is the 3rd Sparda Son
V serves as the third playable protagonist in Devil May Cry 5 and is undoubtedly the most mysterious one since we don't know much about him. The final trailer of the game does hints towards one major fact that has been floating around for quite some time which is that he's the 3rd son of Sparda, which makes him Dante and Vergil's brother.
Now, this scene is hinted at the very end of the trailer when Dante's mother Eva is shown, possibly in a flashback sequence, hiding away a little kid inside a wardrobe and telling him to "change his name and forget his past."
Maybe this is the reason why V's identity has been so much mysterious till now and also his name consists of a single letter because well, he was too young to spell any more letters out? Quite the possibility right?

#2 Vergil confirmed
Vergil - one of the sons of Sparda and brother of Dante has been pretty much teased in the reveal trailer of Devil May Cry 5 itself but this shot at the end of the final trailer just confirms his existence in this game.
Also, he may be seen in two forms - one his normal self and the other in the form of the demon Urizen which Dante, Nero, V, Trish and Lady are seen fighting in the trailer.
#3 Nero is Vergil's son
I think many of us already knew this since Devil May Cry 4 as Nero was shown to inherit devil powers from Sparda and hunt demons just like Dante who also has a demonic arm. Since Dante doesn't have an offspring and the previous game did hint about Vergil's and Nero's relationship, the final trailer just blindly confirms the fact, or at least that's what it seems.
The demon they are fighting named Urizen might be a shadow of Vergil or just another form of him? But at about midway through the trailer, he speaks to Nero saying " You'll regret being born."
Well not something every father tells his son when he's angry but it will do, right? At least in the Devil May Cry universe.

#4 V is dying
This might be the biggest spoiler this trailer provides and I'm totally against it.
V is seen to be in a vulnerable set while Nero tends to him as he speaks" This is my last request." Last request? Seriously? But you're just being introduced into the series.
Well whatever it may be, Capcom did confirm during Microsoft's E3 2018 conference last year that Devil May Cry 5 will move longtime fans of the series to tears and it's going to be a very emotional ride. Prepare yourselves guys.
#5 A throwback to Dante's past
At one point in the trailer, Dante is seen lying in the battlefield, fainted most probably and is dreaming about his mother Eva and his previous home.
The Sparda mansion where Dante and Vergil lived initially is also seen in the trailer with all three of them - V, Nero and Dante exploring and it seems like searching for something. Probably V's connection to the family? Who knows.

#6 The end of the Sparda storyline
Devil May Cry 5 might not be the last Devil May Cry game, but it sure seems to be the last one for the Sparda brothers - that means Dante, Vergil and possibly V.
This surely means that either all of them will die, including Trish and Lady as Nero and Nico continue to the series forward. Nero is the main protagonist of Devil May Cry 5 and Capcom hasn't confirmed that he will have more of screen time than Dante or V.
But the last scene in this final trailer where the screen turns blue and Nero tries to escape from what seems like the place of final battle. It surely means he's the last one standing and the legacy has been transferred.
