On September 29, 2021, just four days after Oleksandr Usyk's victory over Joshua, his promoter Alexander Krassyuk announced that a rematch clause had been activated by Joshua's team. Usyk seemed excited about the prospect of facing Joshua again and expressed his desire for the rematch to take place in Ukraine. However, Joshua's promoter, Eddie Hearn, indicated that the rematch venue was likely to be international or in the UK due to financial considerations.

On June 19, 2022, it was officially announced that the rematch between Usyk and Joshua would take place in Jeddah, Saudi Arabia, on August 20, 2022. This marked Usyk's first defense of his world heavyweight titles, while Joshua aimed to regain his title for the third time.

In the rematch, Usyk defended his belts by securing a split decision victory over Joshua. One judge scored the fight 115–113 in favor of Joshua, while the other two judges scored it 115–113 and 116–112 in favor of Usyk. Notably, Joshua had landed 37 punches to the body compared to 15 in their first encounter. Overall, Usyk performed better in terms of landing punches, with 170 out of 712 compared to Joshua's 124 out of 492. According to CompuBox, Usyk set records for the most punches landed on Joshua in a single round (39 punches in the 10th round).

After the judges' scorecards were announced, Joshua threw two of Usyk's belts out of the ring in frustration. He initially began to leave the arena but returned to the microphone to give an emotional speech about his life, fighting, Ukraine, and praising Usyk. Backstage, Joshua was recorded responding angrily to a bystander's criticism with strong language. During the post-fight press conference, Joshua broke down in tears.
