In July 2018, Mitchell defeated Tyler Diamond at The Ultimate Fighter: Undefeated via majority decision. He continued his success in March 2019 by defeating Bobby Moffett at UFC Fight Night: Thompson vs. Pettis, earning the Fight of the Night award.

In December 2019, Mitchell faced Matt Sayles at UFC on ESPN: Overeem vs. Rozenstruik, securing a first-round twister submission victory, the second in UFC history. He received the Performance of the Night award and 2019 Submission of the Year recognition from various sources, subsequently signing a new four-fight UFC contract.

Mitchell's bout against Charles Rosa, initially scheduled for May 2020, took place on May 9, 2020, at UFC 249, ending in a unanimous decision win for Mitchell with impressive scorecards.

In October 2020, Mitchell fought Andre Fili at UFC Fight Night: Hall vs. Silva, wearing custom trunks. He secured a unanimous decision win.

His victory streak continued in March 2022, defeating Edson Barboza via unanimous decision at UFC 272. A planned main event against Movsar Evloev in November 2022 was canceled due to Evloev's injury.

Mitchell faced Ilia Topuria on December 10, 2022, and lost for the first time in professional MMA via submission in the second round. This was the end of his 15-fight winning streak in MMA.

Scheduled to face Jonathan Pearce in May 2023 at UFC 288, Mitchell withdrew due to a recurring back injury and was replaced by Diego Lopes.
