Spence, who was born on March 3, 1990, in Brentwood, New York, began boxing gym sessions when he was 15 years old and living in Texas. He didn't want to keep going, but his father pushed him to keep going and even switched to the graveyard shift so he could take his son to the gym during the day.

As a result of his victory in the 2009 U.S. National Championship at the Olympic Training Center, Spence Jr. had established himself as the best amateur welterweight in the country. On his way to the London Olympics in 2012, he followed that up with two additional victories in 2010 and 2011. After losing to Andrey Zamkovoy in the quarterfinals, he eventually decided to pursue a professional career.

Spence competed against Chris Algieri, who at the time had a record of 21-2 and had only suffered defeats at the hands of elite competitors Manny Pacquiao and Amir Khan. Spence thoroughly beat Algieri before KO-ing him in the fifth round with a left hook.
