He won the title by beating Sean Strickland via split decision at UFC 297 on January 23, 2024. He is still undefeated in the UFC and has suffered just two losses in his entire professional career so far.

Before his championship win, Du Plessis locked horns with Robert Whittaker at UFC 290. Du Plessis dominated Whittaker and defeated him by second-round TKO to consolidate his position as a top contender for middleweight gold. He was then supposed to face the then middleweight champion, Israel Adesanya, but an injury forced him out, and Adesanya tried to defend his title with Sean Strickland at UFC 294 but ended up losing his title to Strickland via unanimous decision. Strickland tried defending the middleweight title for the first time against him at UFC 297 but failed in his pursuit as Du Plessis won the bout. However, Dricus Du Plessis and Israel Adesanya's fight was confirmed by the UFC and is now scheduled to take place as the main event of UFC 305 on August 18, 2024.
