Jake Matthews was born in Melbourne, Australia, on August 19, 1994. He played football in his younger days, where he represented the Northern Knights for a season. Matthews practiced martial arts to remain fit and trained with his brother under the patronage of his father. He switched to professional mixed martial arts at the age of 18 in 2012. Matthews made his professional mixed martial arts debut on September 15, 2012, against Sam Fiamatai at Shamrock Events: Nights of Mayhem 5 on September 15, 2012, in Melbourne, Australia. He dominated Fiamatai and put his lights off in the second round of the bout to pick up the win by second-round TKO. Matthews went on a smooth winning streak of nine fights before suffering the first loss of his professional career.

Matthews made his debut in the Ultimate Fighting Championship (UFC) while riding his dominating eight-fight winning streak on November 8, 2014. All of his victories had come through finishes, and he locked horns with Dashon Johnson in the first fight for the promotion. Matthews maintained his finishing prowess and finished Johnson in the third round by clutching him in a triangle choke and making him submit. He lost his first professional fight in the UFC while fighting against James Vick in UFC Fight Night: Miocic vs. Hunt on May 10, 2015. Vick defeated him via first-round submission by putting him in a guillotine choke and handing him the first professional loss of his career.
