On September 29, 2021, shortly after Oleksandr Usyk's victory over Joshua, promoter Alexander Krassyuk revealed that a rematch clause had been activated by Joshua's camp. Usyk expressed eagerness for a rematch and a preference for it to be held in Ukraine. However, Eddie Hearn, Joshua's promoter, suggested that the rematch might occur internationally or in the UK due to financial considerations.

Officially, on June 19, 2022, it was announced that the Usyk vs. Joshua rematch would take place in Jeddah, Saudi Arabia, on August 20, 2022. This marked Usyk's initial defense of his world heavyweight title, while Joshua aimed to regain the title for the third time.

In the rematch, Usyk successfully defended his titles through a split decision victory over Joshua. One judge scored 115–113 for Joshua, while the other two judges scored it 115–113 and 116–112 in favor of Usyk. Notably, Joshua landed 37 body punches compared to 15 in their initial encounter. Overall, Usyk outperformed Joshua in landing punches, with 170 out of 712 compared to Joshua's 124 out of 492. CompuBox reported that Usyk set records for landing the most punches on Joshua in a single round, with 39 punches in the 10th round.

Upon hearing the judges' scores, Joshua threw two of Usyk's belts out of the ring in frustration. Initially leaving the arena, he later returned to the microphone to deliver an emotional speech about his life, fighting, and Ukraine and praised Usyk. Backstage, Joshua was caught on camera reacting angrily to a bystander's criticism using strong language. During the post-fight press conference, Joshua broke down in tears.
