DNA Lover episode 12 was released on September 22, 2024, at 9:10 pm (KST) on TV Chosun. The show features Jung In-sun as Han So-jin, a genetic researcher on a mission to find her ideal match, dubbed her "DNA lover," after enduring a series of troubled relationships due to infidelity. Choi Si-won stars as Shim Yeon-woo, a talented obstetrician and the president's son at Shim Hospital.
In DNA Lover, Lee Tae-hwan plays Seo Kang-hoon, So-jin's steadfast friend and a firefighter, while Jung Yoo-jin takes on the role of Jang Mi-eun, a polyamorous romance columnist and one of Yeon-woo's past partners.
In the previous episodes (10 and 11), So-jin discovers Yeon-woo's lies and learns that Kang-hoon is her actual DNA lover. In episode 12, Yeon-woo and So-jin break up, after which Kang-hoon asks her out on a date.

This episode of DNA Lover begins with Kang-hoon dropping So-jin home. After she goes inside, he tries to call Yeon-woo, but his phone is off. Seo-jin struggles to sleep as Yeon-woo's words keep repeating in her head. She wonders how, after sharing his family's issues, Yeon-woo can just leave her like that.
Later, Kang-hoon calls to let her know that he has left some cold medicine for her. So-jin is touched by this gesture, as she knows pharmacies are closed at this time.
The next day in this episode of DNA Lover, Yeon-woo meets So-jin and breaks up with her, returning the DNA bracelet she gave him. She asks why he’s ending things after their deep conversation.
He explains that it is for her future happiness, insisting their connection isn’t enough. He suggests that if she knew Kang-hoon was her true DNA match, she wouldn’t have pursued him.
After Yeon-woo leaves, So-jin is distraught. Meanwhile, a mysterious figure takes photos of her and adds them to a pinboard filled with Yeon-woo’s family pictures.
So-jin tries to forget Yeon-woo with fried chicken and ice cream, but memories flood back. A-ri, seeing her pain, scolds her for having feelings for someone who isn’t her DNA match. In a moment of grief, So-jin takes down Yeon-woo’s pictures and tries to burn them.
Kang-hoon intervenes, thinking she’s playing with fire. Upon hearing about the breakup, he decides to confront Yeon-woo. Yeon-woo insists that his actions are for So-jin’s good, recalling his desire to protect her and his belief that their love wouldn’t last.
The next morning, Kang-hoon invites So-jin to go somewhere with him. That evening, they go jogging in a nearby place, where they talk about some memories they shared. So-jin, however, wonders how she can not smell Kang-hoon's smell.
The next day in episode 11 of DNA Lover, Mi-eun visits Yeon-woo's still unconscious mother. They talk about Yeon-woo and So-jin's breakup. Seeing him sad, Mi-eun asks to take him on a drive, but Yeon-woo declines. She then proceeds to tell him to take care of himself, as now he is taking care of his mother too.
Later in this episode of DNA Lover, Yeon-woo sees photos from the fire scene of his mother's greenhouse that Kang-hoon has sent him. In the picture, Yeon-woo sees a picture of his father with one of his mistresses and remembers meeting her.
The next day, Kang-hoon asks So-ri to help pick him some clothes for a date. After that, with flowers, he goes to meet Han So-jin in her genetic center's auditorium. He has walked in like So-jin's dream from the first episode.
So-jin is surprised to see him and asks about the reason for his being there. Kang-hoon, in reply, asks So-jin for a date to the ice skating ring. Though hesitant at first, So-jin goes with him.
While skating, So-jin sees bubbles, which instantly remind her of her first date with Yeon-woo. She then spots a mailbox with a post sign that brings back memories of a beach day with him. After the date, Kang-hoon drops So-jin home.
Before entering her house, she notices a nearby lamppost in need of repair. Kang-hoon has wrapped a bunch of LED lights and posted an apology note about it. She is surprised by this and remembers a few years ago when they took part in a neurological experiment as a non-couple. The researcher was convinced that they were a couple seeing how the same parts were lightening up in reactions, signifying they felt the same things.
Yeon-woo goes on a blind date next in this episode of DNA Lover, but everything reminds him of So-jin. Mi-eun intervenes, fabricating a story about Yeon-woo’s financial troubles to end the date. The two then have a conversation when Mi-eun reveals that she wants to be with just one person now.
The next day, Yeon-woo is talking to the patient who was trying to jump from the roof of the hospital in the second episode of the drama. He is about to deliver this woman's baby and prepare for her surgery. She asks Yeon-woo to thank So-jin for saving her life and, in the process, her now soon-to-be-born baby.
While in the surgery Yeon-woo keeps remembering the urgency. So-jin saved this lady, asking her to live. After the surgery, he goes to his mother's room and shares with her how even after breaking up, he cannot forget So-jin.
Meanwhile, in this episode of DNA Lover, So-jin is helping Kang-hoon's father at his restaurant while the TV shows the news of a firefighter being stuck in a fire breakout. So-jin and Kang-hoon's father fear it is Kang-hoon, as the description in the news matches with him. They rush to the hospital, but due to the shock, Kang-hoon's father trips on the road.
Coincidentally, Yeon-woo is driving to see So-jin and stops to help them. In the hospital, Kang-hoon's father breaks down crying, seeing Kang-hoon's other colleagues sitting sadly. However, Kang-hoon appears with minor injuries, but another colleague is critically hurt.
Overwhelmed, So-jin embraces Kang-hoon, recalling her father’s death and Kang-hoon being with her through that. Yeon-woo watches from a distance and quietly leaves.
The last scene of DNA Lover reveals the mysterious man to be the priest of A-ri's church. The man goes into the room where Yeon-woo's mother is admitted and unconscious. He is about to take out her oxygen mask, but seeing something, he leaves her. This is where episode 12 of DNA Lover ends.
