- Andrea Navedo as Xiomara "Xo" Gloriana Villeneuva, Jane's mother. She'd become pregnant with her daughter at the age of 16, and Jane seeks to avoid following in her mother's footsteps. She starts out the series as a dance teacher, eager to get into singing.
- Justin Baldoni as Rafael Solano, the owner of the Marbella Hotel where Jane works. He takes an interest in her after her accidental pregnancy, as he's the baby's biological father and is struggling with impotency. He starts out in a fraught relationship with his wife, leading the two to divorce in season 2 after he discovers her infidelity.
- Yael Grobglas as Petra Solano, Rafael's wife who proves to be unfaithful and unscrupulous. After Rafael dumps her for cheating on him, she inseminates herself with his sperm and gives birth to twin daughters. She later finds herself accused of the murder of her twin sister, Anezka, and begins an affair with her defense attorney, Jane Ramos.
- Ivonne Coll as Alba Gloriana Villeneuva, Jane's maternal grandmother who imparts her religious beliefs on her granddaughter and demands Jane save herself for marriage. She was originally an illegal immigrant to the United States until she got her green card.
- Brett Dier as Michael Cordero Jr, a police detective who starts out the series as Jane's husband. Jane's unexpected pregnancy puts him in a love triangle with Rafael, with him initially scheming to keep Rafael married to Petra so that Jane gives away the baby to them.
- Jaime Camil as Rogelio de la Vega, a telenovela star who is also Jane's biological father. Having discovered that he had a child with Xo this entire time, he tries to make amends with her and connect with his erstwhile daughter.
- Anthony Mendez as the Latin Lover Narrator, who narrates over the entire series. The show ends with him being revealed as the grown-up son of Jane and Rafael, named Mateo.
