Yamamoto signed a 12-year deal worth $325 million with the Los Angeles Dodgers on December 27, 2023, along with a $50.625 million posting fee paid to the Orix Buffaloes. He also pitched during the NLCS and World Series, giving up one run in 6⅓ innings in Game 2 of the World Series. He started on the 2025 Opening Day and was awarded the NL Pitcher of the Month for April.
