On April 10, 2011, she was drafted as the third overall pick by the Chicago Sky. She played twelve seasons with the Sky before signing with the New York Liberty prior to the 2023 WNBA season.

Vandersloot attended Gonzaga University, where she played for the Gonzaga Bulldogs women's basketball team. She became a WNBA champion in 2021 and was named to the WNBA All-Rookie Team in 2011. She was a WNBA All-Star in 2011, 2019, and from 2021 to 2023. She additionally led the WNBA in assists in 2014, from 2017 to 2021, and in 2023. Vandersloot was also the WNBA Peak Performer six times in 2015, from 2017 to 2020, and in 2023. In 2023, she also won the WNBA Commissioner's Cup. Vandersloot also participated in leagues in Poland, Hungary, Italy, Russia, and several other countries.
