The center’s basketball journey began at Christ School in Arden, North Carolina, where he led his team to three North Carolina High School Athletic Association state championships. As a senior, Plumlee averaged 15.3 points, 10.1 rebounds, 3.3 assists, and 2.5 blocks per game. He was named the 2009 McDonald's All-American and the North Carolina Mr. Basketball in 2009.
Plumlee then played college basketball for Duke University, where he won the NCAA championship in 2010. In four seasons in college, Plumlee averaged 9.8 points, 7.7 rebounds, and 1.5 assists per game in 141 appearances.
