The Broncos defeated the much-favored Carolina Panthers, who had the best offense in Super Bowl 50, 24–10 in 2016 SB. Miller sacked Panthers quarterback Cam Newton on a third-and-10 play from the 15-yard line in the first quarter. Defensive end Malik Jackson recovered the ball in the end zone for a touchdown, giving the Broncos a 10–0 lead. Since Super Bowl XXVIII, this was the first touchdown from a fumble return in a Super Bowl.
The Broncos were ahead 16-10 with 4:51 remaining in regulation when Miller forced Newton to fumble again, preventing the Panthers' chance to win the game. The Broncos offense then scored a touchdown to complete the victory. Miller was chosen the MVP of the Super Bowl for his effort, which included six tackles, 2.5 sacks, two quarterback hurries, and two forced fumbles. Miller's game at the Super Bowl 50 is believed to be one of the best defensive individual performances in Super Bowl history by many.
In 2022 Super Bowl, with two sacks throughout the game, Miller contributed to the Rams' 23-20 victory over the Bengals in Super Bowl LVI. He is only the second player in Super Bowl history to record two sacks in a single game; the other is New York Giants player Justin Tuck.
Although Miller appeared in three fewer Super Bowls than Charles Haley, he tied Haley's record for the most career Super Bowl sacks with 4.5.
