After an impressive stint with Swift Current in the WHL, the Boston Bruins selected Jake with the 14th overall pick in the 2015 NHL Draft.

Signing a three-year, entry-level contract in 2015, Jake DeBrusk transitioned to the AHL's Providence before earning a spot on the Bruins' roster in 2017. In his debut, he promptly scored his first NHL goal. Throughout his rookie season, DeBrusk established himself as a regular on the second line, tallying 43 points in 70 games. He increased his goal count to 27 in the 2018-19 season, contributing to the Bruins' journey to the Stanley Cup Final, where they faced defeat against the St. Louis Blues in seven games. On the 2022 NHL Trade Day, March 21, 2022, DeBrusk secured a two-year $8 million extension with the Bruins. During the 2022-23 season, he delivered his finest performance, achieving a career-high 50 points and matching his personal best in goals at 27, despite facing setbacks due to injury.

Louie DeBrusk is Jake DeBrusk's father. His father, Louis Dennis DeBrusk, born on March 19, 1971, is a retired Canadian professional ice hockey player. He is currently a color commentator for Edmonton Oilers broadcasts on Sportsnet and the late game of Hockey Night in Canada.

Raised in Port Elgin, Ontario, DeBrusk Sr. began his junior hockey journey with the Port Elgin Bears and Stratford Cullitons before joining the London Knights. Drafted by the New York Rangers in the 1989 NHL Entry Draft, he was later traded to the Edmonton Oilers in a significant deal for Mark Messier. DeBrusk played for various teams, including the Tampa Bay Lightning and the Phoenix Coyotes, accumulating a total of 401 NHL games. Recognized for his fighting prowess, he amassed 1,161 penalty minutes in his career. DeBrusk concluded his professional hockey journey during the 2003–04 AHL season and was inducted into the Cambridge Sports Hall of Fame in 2018.

DeBrusk serves as the primary color analyst for the late game on Hockey Night in Canada. Formerly, he worked as the color commentator for the Phoenix Coyotes radio broadcasts. In 2008, he became the color commentator for NHL on Sportsnet Edmonton Oilers television broadcasts, taking over from Ray Ferraro. Although replaced in 2014, DeBrusk continued as an analyst and later returned to the Oilers broadcast team for the 2020-21 season, partnering with announcer Jack Michaels.

In 2020, Jake DeBrusk, playing in Edmonton for the Bruins, had a unique pre-game interview with his father, Louie DeBrusk, a former Oilers player turned SportsNet broadcaster. The playful interview included jokes about returning to their hometown, hunting stories, and even some banter about hairstyles.
